Our TEM analysis offers an unbiased, in-depth examination of your LNPs. With TEM, you can gain invaluable insights into your product’s overall morphology, particle size, and class distribution, as well as circularity, lamellarity, and membrane thickness. With such high-resolution images, you can ensure that every particle is up to the mark, guaranteeing the safety and efficacy of your final product.

Unveil the True Nature of Your LNPs with High-Resolution TEM Imaging

CryoTEM offers an unfiltered lens into the world of LNPs, accurately illustrating their morphology, size distribution, and the types of particles they contain. In addition, it provides valuable data about the sample's stability or its state of aggregation - all while maintaining the particles in their near-native condition.

But the prowess of CryoTEM doesn't stop there. Its capabilities are further extended to explore the internal density of LNPs. This in-depth examination yields priceless insights into packaging efficiency, revealing the ratio of filled and empty particles in the sample. Alongside, CryoTEM also helps evaluate the signal-to-noise ratio of the image background, providing key information on the sample's purity. Leveraging CryoTEM's potential allows us to offer detailed, unbiased classification and analysis, crucial for the optimization of these therapeutic powerhouses in your pharmaceutical journey.LNPs particle class distribution

Harness the power of detailed analysis with our sophisticated histogram (see figure to the left), which provides an insightful look into the particle class distribution of LNPs as determined through semi-automated detection. The corresponding image to the right then gives you a more tangible perspective, showing the detected and classified particles adorned with easily distinguishable green, blue, and purple outlines.

To provide more context for these classifications:

  • Type 1 LNPs exhibit a homogeneous high internal density, demonstrating a uniform concentration of therapeutic content within the nanoparticle.
  • Type 2 LNPs display a distinct outer shell with a minute internal density, showcasing a strong exterior that's perfect for protecting precious therapeutics during delivery.
  • Type 3 LNPs present a multi-compartmental feature with a heterogeneous internal density distribution, offering a more complex structure that can potentially carry a diverse range of therapeutics.
  • Type 4 LNPs encompass those that don't neatly fit into the above classifications, reflecting the complexity and diversity of these dynamic nanostructures.

FAQ

Can Vironova Bioanalytics provide analysis on filled and empty LNP?

We can provide filled and empty analysis of LNP using cryo-TEM, which allows us to visualize the encapsulant, such as Doxorubicin, inside the LNP. In addition to morphological observations, our analytical report also includes statistical results, such as the ratio of filled and empty LNP.

Can Vironova Bioanalytics perform the stability study of liposomes?

We can perform stability studies of LNP. Contact us to learn more about our stability study services for LNP and to discuss your requirements.

What is the volume and particle concentration of the liposomal product required to perform cryo-TEM analyses?

We require a minimum sample volume of 25 µL and 2 mL of buffer for cryo-TEM analysis, with a recommended lipid concentration of ca. 4 mg/mL for LNP samples.

At which temperatures can Vironova Bioanalytics store LNP samples?

We offer sample storage at four different temperatures: -86°C, -20°C, +4°C, and room temperature.
